Install the electrode. Check the connections before sterilization, whether the appearance of each pipeline is normal, and whether the steam outlet pipe is firm. Each valve should be checked by two persons to see if it is fully closed.

Sterilization

At least two persons are required to be present during sterilization. One person is responsible for operating various valves, and the other person is responsible for observing and reporting various parameters.

After confirming that all valves are closed, open the steam generator inlet valve, start the steam generator, and the steam generator starts to automatically enter the water. At this time, the drain valve at the lower part of the steam generator should be opened, and the drain valve should be closed after the rust is removed as much as possible. . Pay attention to observe the water level in the steam generator to prevent accidents.

At this time, fully open the jacket air outlet valve, slowly open the steam generator main valve, slowly partially open the jacket air inlet valve, and start to introduce water vapor into the jacket. At the beginning, the steam pressure may be unstable, and the pointer of the pressure gauge may swing violently. You can properly close the small jacket inlet valve, and then open the sampling valve steam valve, the sampling valve steam valve, the bottom valve steam valve, and the bottom valve steam valve. , Sterilize the sampling valve and the bottom valve, and drain out the water vapor with unstable pressure.

When the water vapor pressure does not change drastically, close the valve and the outlet valve in turn. The temperature in the tank gradually rises, and when the set temperature is 90 °, the stirring motor stops rotating. At this time, the tank main air inlet valve is opened, the tank body air outlet valve is opened, and the following valves are opened in sequence: open the air outlet adjustment valve, and slowly open the tank body The steam inlet valve feeds steam into the tank, and the air filter gauge pressure starts to rise. When it exceeds 0.1MP, the condensate valve is opened to drain, and the large tank body steam inlet valve continues to be opened. After the tank air is exhausted, the tank internal pressure slowly rises and adjusts The gas outlet regulating valve can reach the temperature and pressure required for sterilization. When the sterilization temperature is reached, the timer starts.

Adjust the jacket inlet valve, tank inlet steam valve and outlet regulator valve to keep the temperature and tank pressure at the set temperature. After the sterilization time expires, close the opened steam inlet valves in turn, close the steam main valve, completely release the exhaust gas exhaust valve in the tank to release the pressure in the tank, and turn off the power supply of the steam generator. Turn on the air compressor. When the tank pressure drops to 0.05MP, slowly open the intake air adjustment valve to maintain the positive pressure in the tank.

During the sterilization process, the operator should wear long trousers and protective gloves to prevent burns. If the steam generator automatically feeds water during the sterilization process, the total steam pressure will drop rapidly. If necessary, the steam main valve can be opened appropriately to maintain the pressure, but be careful to observe the pressure change. When steam is passed into the tank, the tank pressure drops faster after sterilization, pay attention to the timing of passing sterile air. Try not to make the air compressor and steam generator work at the same time. The air filter pressure should not exceed 0.2MP during the sterilization process. The size of the condensate will affect the concentration of the liquid in the tank after sterilization, and the increase in steam condensate should be considered.

Fermentation

By controlling the size of the intake valve and exhaust valve, the ventilation volume and tank pressure can be adjusted, and the stirring rotation speed can be controlled so that the fermentation dissolved oxygen meets the process requirements. During fermentation, the tank main water inlet valve is opened, and the jacket outlet valve and jacket inlet valve are opened. At this time, the system automatically controls the fermentation temperature by controlling the jacket water temperature.

Due to the positive pressure in the tank, close the small intake valve when inoculating, reduce the tank pressure appropriately to 0.01-0.02MP, use the flame ring protection to quickly open the inoculation port (to prevent the air flow from extinguishing the flame), and quickly close the inoculation port after inoculation. Sterilize the bottles and auxiliary hoses of the feeding system into the host (note the direction), pierce the silicone pad under the protection of the flame ring, and install the feeding system. At this time, real-time monitoring of various parameters of the system should be displayed on the workstation. Pay attention to parameter recording.

When the fermentation progresses to the later stage, there may be bacteria that block the trachea. The phenomenon is that the ventilation volume decreases and the tank pressure slightly increases. At this time, manually turn the air outlet valve left and right to ease the blockage.
