Labeled DNA probe

If you have a sufficient amount of high-purity templates, it is recommended that you use the random primer labeling method of DIG High Prime DNA Labeling and Detection Starter Kit I and II. These two kits contain all the reagents required for labeling, blocking, hybridization and detection, and are truly one-stop service kits. The difference between the two lies in the different detection methods: Starter Kit I uses the color development method and uses the color development substrate NBT / BCIP; while Starter Kit II uses the chemiluminescence method CSPD.

If the amount of DNA template used to synthesize the probe is relatively small or the quality is not high enough, it is recommended that you use the PCR DIG Probe Synthesis Kit. This kit is dedicated to preparing highly sensitive hybridization probes, such as single copy genes. Another feature is that the quantitative spot detection process is not required in the experiment, and the efficiency of PCR labeled probes can be quickly determined by quantitative gel electrophoresis.

DNA probes labeled with a gap translation method can control the length of the probe, and are generally used for in situ hybridization.

Labeled RNA probe

In Northern blot, either DNA probes or RNA probes can be used. Generally, the specificity and sensitivity of RNA probes will be higher; of course, if there are no particularly strict requirements on sensitivity and specificity, DNA probes will be more convenient to operate.

If you need to prepare RNA probes, it is recommended that you use the Northern Starter Kit. The kit uses linear DNA as a template and incorporates DIG-11-UTP into the transcribed RNA through the action of SP6, T7 or T3 RNA polymerase, which is detected by the chemiluminescent substrate CDP-Star. Another product, DIG RNA Labeling Kit, can also be used for RNA probe labeling.

Oligonucleotide
If your probe template is very short and the amount is relatively small, you can choose the end labeling method to label the oligonucleotide probe.
