Rat osteocalcin (OC) Elisa kit

The kit employs a double-antibody sandwich immunoassay to quantify the level of osteocalcin (OC) in the sample. A microwell plate is pre-coated with a purified rat-specific OC antibody, creating a solid-phase capture antibody. The sample containing OC is then added to the wells, allowing the OC to bind to the immobilized antibody. Following a washing step, an HRP-conjugated OC antibody is introduced, which binds to the captured OC, forming a complex of antibody-antigen-enzyme-labeled antibody. After thorough washing to remove unbound components, the substrate TMB is added. Under the catalytic action of HRP, TMB turns blue, and upon addition of an acidic stop solution, it changes to yellow. The intensity of the yellow color is directly proportional to the concentration of OC in the sample. The optical density (OD) is measured at 450 nm using a microplate reader. By comparing the OD values against a standard curve generated from known concentrations of OC, the exact concentration of rat OC in the sample can be accurately determined. This method ensures high specificity and sensitivity for OC detection, making it ideal for research and diagnostic applications.
