1. Principle:
The decomposition of glucose by bacteria is divided into oxidation type and fermentation type.
Oxidizing bacteria can only decompose glucose under aerobic conditions, and cannot decompose glucose under anaerobic conditions; fermenting bacteria can decompose glucose under aerobic and anaerobic conditions;
Bacteria that do not break down glucose are called alkali-generating. According to the color change of sugar tube, bacteria can be identified. 2. Method: Take the bacteria to be tested and inoculate it on two tubes of glucose semi-solid medium by puncture method. Add a layer (about 1cm) of sterilized liquid paraffin to one tube to isolate the air and incubate at 37 ℃ for 2 ~ 4h Every day, observe the results every day.
Application: It can be used to distinguish Enterobacteriaceae bacteria (fermentation type) from non-fermentation bacteria (oxidation type or alkali-producing type). It can also be used to identify staphylococci (fermentation type) and micrococcus (oxidation type).
