GE launches new affinity chromatography medium CaptoL

GE Healthcare recently launched a new affinity chromatography medium Capto L, which is suitable for the capture of antibodies and antibody fragments. It combines a rigid, high-flow agarose matrix with a recombinant protein L ligand that binds to immunoglobulin. This ligand has a strong affinity for the variable region on the antibody Kappa light chain. Therefore, Capto L is suitable for capturing various antibody fragments, such as Fab, single chain antibodies (scFv) and domain antibodies (Dabs).

Monoclonal antibodies are usually purified using Protein A affinity chromatography. However, corresponding purification schemes for antibody fragments have been lacking. One of the reasons is the high diversity of antibody fragments, and the other reason is that the chromatography media currently available on the market do not meet the requirements of industrial-scale purification. The introduction of Capto L provides a basis for the purification platform method of this biomolecule category.

Compared with ion exchange and hydrophobic interaction chromatography, the affinity chromatography capture step has a very stable and versatile process. The optimization limit required for purification with Capto L is minimal, and higher purity can be obtained in one step. This obviously saves the cost, time and efficiency in the process. Compared with other methods, because the high selectivity of the affinity chromatography capture step will help to reduce the subsequent fine purification step, less selective purification media is used in production, which can also benefit from it.

With its highly dynamic binding capacity and high selectivity, Capto L improves the process economy and process design flexibility.
